BPC-157

BPC-157 is a synthetic 15-amino-acid peptide derived from a sequence in a human gastric juice protein. Research consists almost entirely of rodent and cell studies, with very limited human data [1][2].

Evidence: preclinical · Mostly rodent and cell studies

Where it’s been studied

1234
  • 1TendonAchilles detachment model, rat (2006); tendon fibroblasts, rat cells (2014)
  • 2Blood vesselsHind-limb ischemia model, rat; endothelial cells (2017)
  • 3GutIntestinal anastomosis models, rat (review, 2024)
  • 4CellularHuman vascular endothelial cells and rat tendon fibroblasts (2014, 2017)

Mechanism studied

In endothelial cell and rat studies, BPC-157 increased VEGFR2 expression and internalization and activated the VEGFR2-Akt-eNOS signaling pathway, which researchers linked to increased vessel formation [3]. In cultured rat tendon fibroblasts it increased growth hormone receptor expression, and added growth hormone then activated JAK2 signaling and cell proliferation [5]. Interactions with the nitric oxide system and other pathways have been proposed from rodent work but are not established [1][2].

What the research shows

StudyModelReported resultSource
Achilles detachment model [4]Rat
Achilles tendon surgically detached from bone, rats; with and without corticosteroid
Researchers measured higher load to failure, stiffness and Young's modulus, more organized collagen fibers, and higher collagen type I than controls.Peer-reviewed
J Orthop Res 2006
VEGFR2 and vessel formation [3]Rat
Hind-limb ischemia model in rats, plus chick embryo membrane assay and human endothelial cell culture
Faster blood-flow recovery on laser Doppler, more vessels on histology, higher VEGFR2 mRNA and protein; VEGF-A unchanged in cells.Peer-reviewed
J Mol Med 2017
Growth hormone receptor in tendon cells [5]Cell
Rat Achilles tendon fibroblasts in culture
Growth hormone receptor mRNA and protein increased in a dose- and time-dependent way; added growth hormone then increased proliferation and JAK2 activation.Peer-reviewed
Molecules 2014
Pharmacokinetics and excretion [6]Other animal
Rats and beagle dogs, single and repeated dosing, radiolabeled tracing
Elimination half-life under 30 minutes in both species (15.2 min rat, 5.27 min dog); broken down into small fragments and amino acids, mainly proline.Peer-reviewed

Intestinal anastomosis research [7]Review
Review of rat studies of surgical intestinal, nerve and vessel anastomoses and fistula models
