Gonadorelin

Also known as: GnRH

Gonadorelin is a synthetic decapeptide with the same amino acid sequence as natural gonadotropin-releasing hormone (GnRH). It has a long research record, mostly in cattle reproduction studies and human pituitary-function research.

Evidence: moderate · Large cattle studies; human data mostly small or retrospective

At a glance

Class
Synthetic gonadotropin-releasing hormone
Developer
Originally developed as a pharmaceutical; approved veterinary products exist
FDA status
Human brand (Factrel) was FDA-approved and is discontinued in the US; veterinary gonadorelin products are FDA-approved for cattle. This page covers the research compound only.
Human data

Animal / lab data
Randomized studies in dairy and Bos indicus cattle (up to 558 animals)

Where it’s been studied

12
  • 1PituitaryLH release, lactating dairy cows (2009) and Bos indicus cattle (2022, 2024)
  • 2ReproductiveOvulation response, cattle (2009, 2024); adult men, retrospective (2025)

Mechanism studied

Gonadorelin has the same 10-amino-acid sequence as hypothalamic GnRH (pGlu-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH2) and acts on pituitary GnRH receptors to trigger release of LH and FSH [1]. In cattle, researchers measured a rise in serum LH within about 1.35 hours of administration [4]. Because it is the native sequence, it has been compared with longer-acting synthetic GnRH analogs such as buserelin, which produced higher LH peaks [3].

What the research shows

StudyModelReported resultSource
Comparison of four gonadorelin products [2]Other animal
Lactating dairy cows, randomized to four commercial gonadorelin products (n=132-146 per group; 558 total)
Ovulation of an accessory follicle ranged from 55.3% to 85.0% across products; LH peak, timing and area under the curve did not differ between products.Peer-reviewed
Theriogenology 2009
Gonadorelin vs buserelin in Bos indicus cattle [3]Other animal
Nelore heifers (n=57) and cows (n=53) on day 7 after a prior ovulation; two gonadorelin and two buserelin amounts
Buserelin produced higher LH peaks than gonadorelin; in heifers, ovulation was 88.9% with buserelin vs 16.7% with gonadorelin.Peer-reviewed
Theriogenology 2024
Two gonadorelin amounts vs saline [4]Other animal
Bos indicus heifers and cows (n=114), randomized: untreated, saline, or one of two gonadorelin amounts
Both gonadorelin groups had higher serum LH than saline (P<0.001); new corpus luteum formation by day 5 (about 14%) did not differ from saline (12.5%).Peer-reviewed
Aust Vet J 2022
Pulsatile GnRH cohort in men [5]Human
Adult men, retrospective, single center, no control group (n=54); mean follow-up 15.9 months
Endpoints: reproductive-axis hormone measures over 24 months. Published.Peer-reviewed
