Mazdutide is a synthetic once-weekly peptide that activates both the GLP-1 and glucagon receptors. It has been studied in multiple phase 1 to phase 3 human trials, mostly in China, plus a small body of animal and tissue work.
Mazdutide is a peptide agonist at both the GLP-1 receptor and the glucagon receptor [1][3]. In isolated human atrial tissue, its effect on contractile force was reduced by both a GLP-1 receptor blocker and a glucagon receptor blocker, consistent with action at both receptors in that tissue [6]. Hippocampal gene-expression and synaptic-marker changes seen in db/db mice are reported findings; whether mazdutide crosses the blood-brain barrier has not been established [5].
