5-Amino-1MQ

Also known as: NNMT inhibitor (small molecule)

A small molecule (not a peptide) from the methylquinolinium family that blocks the enzyme nicotinamide N-methyltransferase (NNMT). Research is limited to a handful of mouse and cell studies from one academic group.

Mechanism studied

5-amino-1MQ blocks NNMT, an enzyme that uses up nicotinamide and the methyl donor SAM to make 1-methylnicotinamide [1]. In cultured fat cells, blocking NNMT lowered 1-methylnicotinamide and raised NAD+ and SAM, and it did not inhibit related methyltransferases or NAD+ salvage enzymes [1]. Researchers propose that the shift in NAD+ balance underlies the fat-cell and muscle-stem-cell effects seen in mice; this remains a hypothesis [2].
