An investigational long-acting analog of the pancreatic hormone amylin, developed by Zealand Pharma with Roche. Human data so far come from phase 1 and phase 2 trials reported at conferences and in company releases; rat data come from conference posters.
Petrelintide is an engineered analog of amylin, a hormone released from pancreatic beta cells together with insulin after meals; it is built for once-weekly use and, per the developer, stays stable without forming fibrils at neutral pH [1]. The developer proposes action at amylin receptors with possible effects on leptin signaling, a mechanism described as distinct from GLP-1 receptor agonists [1]. This proposed mechanism has not yet been detailed in a peer-reviewed paper.
| Study | Model | Reported result | Source |
|---|---|---|---|
| ZUPREME-1, phase 2 [2] | Human 493 adults, randomized, double-blind, placebo-controlled, five levels vs placebo, 42 weeks plus follow-up (NCT06662539) | Primary endpoint: prespecified metabolic measure at week 28. Presented at ADA 2026; not yet published in a journal | Company-reported Zealand Pharma press release 2026 |
| Phase 1 single and multiple administration [3] | Human Single-administration part: 64 healthy men; multiple-administration part: 20 healthy men, 6 weeks; randomized vs placebo | Measured tolerability, gastrointestinal adverse events, steady-state pharmacokinetics and body weight change. Conference poster | Conference abstract ADA Scientific Sessions 2024 (1668-P) |
| Body composition study [4] | Rat High-fat-diet model, rats, n=10 per group, 30 days, vs vehicle and liraglutide | Researchers reported body-weight and body-composition measurements vs vehicle and liraglutide; see poster. | Conference abstract ADA Scientific Sessions 2024 (1662-P) |
| Food choice study [5] | Rat High-fat-diet model, rats with free access to chow and high-fat diet, n=10 per group, 30 days | Researchers measured high-fat-diet and chow intake vs vehicle; chow intake unchanged in all groups. | Conference abstract ADA Scientific Sessions 2024 (1661-P) |
